Fill a coplin jar with sufficient Antigen Retrieval Solution (see Step 2) to cover the tissue sections on the slides. Prepare a 1X Antigen Retrieval Solution by mixing 10 mL of Antigen Retrieval Reagent, pH 6 (10X) with 90 mL of deionized water to make 100 mL of solution. The disadvantages of PIER are the low success rate for restoring immunoreactivity and the potential for destroying both tissue morphology and the antigen of interest. HIER is the most common approach to antigen retrieval, and the temperature, pH and time of incubation are critical factors that must be optimized for proper antigen unmasking without causing morphological damage.
